Is this a reverse rotation pump?

Yes, all waterpumps for the Jeep inline 6cyl are reverse rotation (It turns counterclockwise looking from the front), So few water pumps turn clockwise that reverse rotation is really standard rotatio, if you look closely at the photo of the impeller you will see the letter "R" stamped on it.

What size is the threaded hole that a hose would go to the intake or heater from?

3/4 inch standard. DORMAN makes that replacement part
