How much does this tin cover?

Hi, the tin should cover between 3.7m2 and 5.2m2

Can it be used on wood and can it be applied to a vertical surface

It can certainly be applied to a vertical surface I think it's good on pretty much anything

I need to waterproof an area around a glass skylight set in a metal frame will this product combined with your liquid rubber product do the job?

I'm sorry to say, I have no idea, this is a question that you need to ask the makers of Isoflex. I'm going to use it for sealing a water tank on my model steam engine, which I am making. Sorry I couldn't be of much help. If you read some of the comments for Isoflex, there might be some help there.
